Sugar Chip

Sugar Chip is an array of structurally defined sugar chains (carbohydrates) immobilized on a thin gold chip.
One type of sugar or dozens of different sugars can be immobilized on a single chip. The unbound portion of the sugar chain is free to interact with sugar-binding proteins such as lectins.
How does the Sugar Chips and Surface Plasmon Resonance (SPR) work? Light is focused on the opposite side of the Chip from where the sugars are immobilized.
As light passes through the chip, it excites the surface plasmon, the oscillations of free electrons propagating along the chip' s surface. The angle at which the light is reflected off the surface is called the SPR angle.
When lectins bind to the immobilized sugars, the surface plasmon on the opposite side of the chip is altered. This is directly proportional to the change in bound mass. Real time measuring of the SPR angle reflects changes to the surface concentration or amount of bound lectins.
Together, Sugar Chips and SPR technology can detect real-time biological interactions between carbohydrates and carbohydrate-binding proteins without labeling target proteins.
1. Screening analysis with SPR imaging
Conduct a holistic analysis of protein-sugar interactions, by immobilizing dozens of sugar-chains on a single Sugar Chip array for SPR imaging.
This serviceis ideal for screening sugar-chain binding proteins in cell or acterial lysates.
In SPR imaging, binding interactions between sugar chains and proteins is observed by spot intensity.

2. Kinetic analysis of carbohydrate - protein interactions
Characterize protein specificity and affinity using SPR and a Sugar Chip spotted with one type of sugar.
Kinetic analysis can be used as a stand alone custom service or as a follow up service to the screening analysis for additional lectin characterization.

SGNP

SGNP(Sugar chain - immobilized Gold Nano-Particle) is a gold nano particles immobilized with structurally defined sugar chains, showing red-purple color (lmax = ca. 530 nm).
SGNP is a very convenient tool for evaluating sugar chain - protein interaction, since the interaction can be detected visually, and can be quantified from the change of OD at 530 nm.

There are several applications as follows:
1. Aggregation of protein
2. Inhibition assay
3. Dot-blotting
4. Isolation and purification of lectin from a crude extract
*SGNP is a reagent for research. Do not use for other purposes.

Application#1: Aggregation assay of protein(s)
Regularly sugar chain binding proteins, such as lectins, possess multiple binding sites for sugar chains. By the addition of SGNP to the protein solution, the protein may form aggregates with sugar chains immobilized SGNP in a short time of period. The change can be seen visually, or can be quantified by measuring OD at around 530 nm. Using SGNP, the binding properties (selectivity, dissociation constant (K D), specificity, etc.) are easily evaluated.

Application#2: Probe for Dot-Blotting assay
The sugar chain binding potency of the trace amount of your samples can be detected.

Application#3: Isolation and identification of target protein from a crude extract
Quick isolation and identification of protein(s) having multi-binding sites (lectin) from a plant-derived crude extract of cell-lysate are available using SGNP. Once you get an aggregate by mixing appropriate SGNP with your extract or lysate, the aggregate can be directly applied for SDS-PAGE. Then, the protein band(s) in SDS-gel is analyzed according to the proteomics procedure.
